Killie's U20s were edged out by Hamilton in a closely contested match at Rugby Park.
A single goal decided the match with Jasko Keranovic unable to keep the ball out following a looping clearance from an Accies corner.
Andy Millen stuck with the bulk of the side following last week's victory over Aberdeen with Adam Frizzell coming into the team.
Frizzell was Killie's main creative spark throughout, orchestrating many attacks and seeing a fine effort rattle off the cross bar after the break.
Ross Cunningham impressed for the visitors, showing excellent close control as well as strong positional play to test Keranovic on a number of occasions.
After appearing as a late substitute in last night's first team win over Dundee, Aaron Simpson started at right back and he got a full 90 minutes under his belt.
Innes Cameron came into the match with two goals in as many matches in 2018 but despite another strong showing he was unable to find the back of the net this time around.
Accies manager Martin Canning directed his team from the sidelines as the match opened up in the second period but Killie could not find a clear route to goal as the visitors came out with all three points.
Kilmarnock U20s: Keranovic; Simpson, Waters, Dempsie, Spencer (McLevy), Mouanda, Scally (McDonald), Frizzell, Cameron, Samizadeh, Graham (Paterson)
